It is generally known that etiologic factors for esophageal cancer include genetic susceptibility, alcohol intake, tobacco smoke and diet constituents. The results from hospital-based case control study in southern Thailand indicated that a strong association might exist between esophageal cancer and frequent consumption of the local bean, Archidendron jiringa. However, little is known about its mutagenicity or carcinogenicity. To investigate this question, we examined the promoting activity of Archidendron jiringa using diethylnitrosamine (DENA) induced preneoplastic foci as end point markers in rat liver receiving a 10% bean diet. For initiation, three week old rats were given 50mg/kg of DENA intrapenitoneally 2 times at a 2 week interval. No toxicity was observed in the group receiving 10% bean diet alone. However, the number of GST-P positive foci were increased significantly in the supplemented as compared with the DENA alone group. This is the first demonstration that the local bean Archidendron jiringain, can acts as a promoter of DENA-induction of preneoplastic foci in rat liver.
