Experimental Verification of the Concepts of Ovarian Tumor Classification: Takashi NISHIDA, Toru SUGIYAMA, Seisuke KUMAGAI, Nobuyuki HIRAI, Michiaki YAKUSHIJI (Dept. of OB/GY Kurume Univ.)

[Background] The surface epithelium of the human ovary simulates the mullerian form in tumor formation. In experimental animals, however, such a phenomenon is not known. [Method] A chemical carcinogen, 7,12-dimethlybenz[a]anthracene (DMBA) was heated, and the central portion od a 3-0 silk suture was immursed in the melted caricnogen. The DMBA-coated silk was insterted in the ovary 40 7-weeks old Wistar stain rats following the vaginal cytology. Animals were sacrificed when the tumor mass became large enough to extend the abdominal wall. The exprimental period lasted for 60 weeks. THe tumoe histologies were compared to those in the human counterpart and the mullerian derivatives in rat. [Results] In ninteen rats, including 4 animals with cornified vaginal smear at the time of DMBA-treatment, an ovariancancer developed within 36 weeks. In 17 epitherila tumors the neoplastic cells proliferated to form papillary or glandular structures, and the lining epithelium consisited of flattend or cuboisal cells and columnar cells, often with pseudostratified nuclei. Adenocarcinoma components transformed to squamous cells in 2 cases. The remaining 2 neoplasms were to pure mesenchymal tumors manifesting a histology of fibrosarcomatous tumor, and one of them contained heterologous malignant osteoid component. In the remaining 21 rats, including 5 animals with cornified smaer, no tumors developed during the experimental period. [Conclusion] Our results suggest that the surface epitherium of rat ovary also simulates the mullarian form in tumor formation.
