Recent studies have indicated that the t(12;22)(q13;q12) translocation and EWS/ATF-1 fusion gene induced by this translocation are pathognomonic for clear cell sarcoma. Only a few clear cell sarcoma cell lines have been reported, but they have not been fully characterized at the molecular level.
We reported the establishment and morphological features of a clear cell sarcoma cell line, HS-MM, in 1993. In the present study, this cell line, maintained for more than four years since establishment, was further characterized by light microscopy, cytogenetical studies, fluorescence in situ hybridization (FISH) analysis and reverse transcriptase polymerase chain reaction (RT-PCR). HS-MM cells both in vitro and in vivo revealed essentially the same morphological features as clear cell sarcoma cells, exhibiting positive immunoreactions for S-100 protein and HMB45. In addition, they exhibited pseudodiploid karyotypes with the specific t(12; 22)(q13;q12) translocation. The translocation between chromosomes 12 and 22 was confirmed by FISH analysis, and the hybrid EWS/ATF-1 transcript was detected by RT-PCR.
The HS-MM cell line will be extremely useful for further studies of clear cell sarcoma.
