HTLV-I 産生細胞との混合培養により樹立された胸腺由来 myogenic progenitor細胞株の生物学的性状:岡 剛史、林一彦、赤木忠厚、(岡山大・医・第二病理)

Characterization of thymus-derived myogenic progenitor cell line, ST1, established by the co-culture with HTLV-I producer cells: Takashi OKA, Kazuhiko HAYASHI, Tadaatsu AKAGI (Dept. Pathology., Okayama Univ. Med. Sch.)

We established thymus-derived myogenic progenitor cell line (ST1) which can be induced to differentiate to myoid cells in vitro, by the co-cultivation of rat whole thymus cells with lethally X-ray irradiated HTLV-I producer cell line MT2. This cell line shows epithelial cell like morphology and rapidly grow in the normal culture condition ;10% FCS, DMEM. However, about one week after changing to growth arrest condition in the presence of 1% FCS, cells became larger and formed myotube-like large multinuclear cells and started to contract spontaneously. This differentiated cells expressed smooth muscle actin, myosin, myoglobulin, acetyl chorine receptor, vimentin, desmin and S100 antigens. Progenitor cells were negative for these antigens. Electron microscopic study shows immature Z-band-like structure and "honey-comblike" tubular array in the cytoplasm. HTLV-I genome was not detected in this cell line with Southern blot analysis, suggesting cytokines or lymphokines secreted by X-ray irradiated MT2 cells may supported to grow this myogenic progenitor cells  and  induced adaptation to in vitro culture condition to become cell line. Thymic myoid cells have been reported to increase the cell number in the case of myasthenia gravis and thymoma. This cell line will be useful for biologic and pathologic investigation on these diseases.
