大腸癌におけるGlucose transporterの発現

We conducted this study to elucidate the expression of five facilitative glucose transporters in colorectal carcinoma, normal colonic mucosa and adenoma. Materials and Methods: Expression of GLUT protein was examined by immuno-histochemistry in 35 colorectal carcinoma(25 invasive carcinoma, 11 cancer in adenoma), 30 adenoma and 10 hyperplastic polyps. Total RNA was extracted from 13 specimens of colorectal carcinoma and expression of Glut mRNA were investigated by RT-PCR. Results: Immuno-histochemically, 62% of colorectal carcinoma was positive for anti-Glut1 antibody, 44% was positive for Glut 2. Glut 3, 4 and 5 were positive in 26%, 22% and 45%of cases, respectively. Expression of Glut1 in adenoma and hyperplasia was less frequent than that of carcinoma, 13% and 10% respectively(p<0.05). Glut4 was rarely expressed either in adenoma or hyperplasia (p<0.05 vs. Carcinoma). Normal mucosa was not stained with all immunostaining. Glut1 mRNA was expressed in 5 cases, Glut2 mRNA in 11 cases, Glut3 mRNA in 4 cases, Glut4 mRNA in 11 cases, and Glut5 mRNA in 2 cases, respectively.
