Osteopontin(OPN), RGD-containing glycoprotein has been suggested to function in adhesive interactions at the tumor-host interface and influence tumor invasion and metastasis. A monoclonal antibody was developed to synthetic peptide representing K170 through E187 sequences of human OPN,and used for ABC immunostaing. Formalin-fixed and paraffin-embedded human tumor tissues including 27 carcinomas(3 lung,1 tongue,2 esophagus,4 stomach,7 colon,9 breast,1 ovary),1 duodenal sarcoma,1 glioma,and 1 meningioma were examined. Macrophage-like cells were consistently positive in all tissues examined. In 16 tumors(53.3%),tumor cells in certain areas particularly necrosis and degenerating cells were positive. These results demonstrated OPN expression by macrophage-like cells and some tumor cells in formalin-fixed and paraffin-embedded human tumor tissues. A possible significance of OPN in tumor cells requires further study.
