Effect of ginseng on N, N'-dimethylhydrazine - induced aberrant crypt foci in the colon of rats : Wei LI, Hideki WANIBUCHI, Elsayed I. SALIM, Shinji YAMAMOTO, Shoji FUKUSHIMA ( Department of Pathology, Med.Sch.Osaka City Univ.)

   Ginseng has been proposed for prevention of certain types of cancer in animal experiments. Colorectal cancer is a major target for chemoprevention because it remains the third most malignant neoplasm in the world. The modifying effect of dietary administration of ginseng on the development of N, N'-dimethylhydrazine (DMH)-induced colonic aberrant crypt foci (ACF), putative preneoplastic lesions,was investigated in rats. Fifty male 6-week-old F344 rats were given s.c.injections of DMH (20 mg/kg body wt.) once a week for 4 weeks for colon carcinogenesis. They also received diets containing white ginseng or red ginseng for 5 weeks, starting 1 week before the first dose of DMH, or for 8 weeks after DMH treatment. At termination of the study (week 12 ) dietary administration of red ginseng together with DMH, was found to cause a significant reduction in the frequency of the total numbers of ACF. Expression of cell proliferation biomarker 5-bromo-2'-deoxyuridine (BrdU) labeling-index in colonic epithelium was reduced in animals that received red ginseng simultaneous DMH administration. These findings indicate that red ginseng might exert a preventive effects on DMH induced colon carcinogenesis in rats, which would likely lead to the prevention of colon cancer.
