Selaginella tamariscina (ST), an oriental medicine, is known to exert tumorlytic effects. In this study, ST was extracted using water and several organic solvents, and then each fraction was assayed for its anti-tumoral effects by 3-(4,5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide (MTT). Among the effective fractions, the water-extracted fraction proved to be efficient to increase the expression of p53 tumor suppressor gene when evaluated by northern blotting method and to induce G1 arrest in cell cycle when analyzed by flow cytometry. The modifying effects of pulverized ST on cell proliferation in the stomach were investigated in rats treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). Male 5-week-old Wistar rats were given by gavage 150 mg/kg of MNNG dissolved in dimethyl sulfoxide and then respectively fed a diet supplemented with 5%, 1% or 0% of ST from 6h after the MNNG treatment for 42h. The 1% ST feeding caused a significant reduction (p<0.05) in proliferating cell nuclear antigen(PCNA) -labeling index in the glandular stomach epithelium as compared with the MNNG alone group value although the 5% ST feeding only showed a tendency for decrease. These results suggest that ST could be a chemopreventive agent against gastric cancer.
