The expression of c-Ret protein, a product of c-ret proto-oncogene, was studied in thymomas developed spontaneously in old BUF/Mna rats and in normal thymuses of young BUF/Mna and ACI rats. In immunostaining, the expression of c-Ret protein was found in epithelial cells, in which cytokeratin was contained, of the medulla in the nomral thymuses, but it scattered in all over the areas of the thymomas. Western blot analysis showed that the c-Ret protein was of mainly 170-kDa in the normal thymuses, but of 150-kDa and 170-kDa in the thymomas. It is known that the 170-kDa c-Ret protein represents a mature glycosylated form present on cell surface and the 150-kDa c-Ret protein is an immature glycosylated form in the cytoplasm. An immature 150-kDa c-Ret protein appeared in the thymomas, but not in normal thymuses, may be resulted from either mutation of c-ret proto-oncogene, or the disfunction of glycosidase in the processes of condensation and transportation of c-Ret in the cytoplasm.
