Notchシグナルに関わるhiiragi (hrg)遺伝子の単離:村田 武英, 横山 和尚(理研筑波センター)

Identification of hiiragi (hrg) gene that associates with the Notch signalling: T. Murata, K. K. Yokoyama (Tsukuba Life Science Center, The Institute of Physical and Chemical Research (RIKEN), Tsukuba, Ibaraki 305, Japan)

The region specific expression of genes involved in the development owes specification of compartment and its boundary. The fate of each part of adult wing of Drosophila melanogaster has been studied and genes that responsible for the formation of the dorsal/ventral compartment boundary have been identified. cut, wingless and vestigial are known as such the genes and the boundary specific expression of these genes is regulated by one of the cell-cell signalling, Notch. We have identified a new gene of the fruit fly that associates with the signal cascade of Notch, known as int-3 of mouse and TAN-1 of human. A loss of function mutation of hrg has notched wing phenotype and enhanced loss of function mutation of Notch and deltex. The genetic interaction between hrg and the members of Notch signal cascade suggests that hrg act as a positive factor in the Notch signalling. Here we show further evidences of involvement of hrg in the Notch signalling: (1) Outgrowth of adult wing was affected by the hrg mutation and (2) ectopic expression of cut by the forced expression of Notch and Serrate, a ligand for Notch receptor, was reduced in a hrg mutant.
