c-Ras and the Fokal Adhesion Kinase (FAK) are required for the activation of Mitogen-activated protein kinase (MAPK) by Hyaluronic Acid in 3Y1 cells: Mihaela SERBULEA1, Michinari HAMAGUCHI2 (1Dept. of Internal Medicine III,2 Dept. of Molecular Pathogenesis, Nagoya Univ.School of Medicine)

Hyaluronic Acid (HA) is known to be implicated in cell proliferation, differentiation and locomotion. Despite its biological importance, the intracellular signaling pathway remains largely unknown. Here we show that HA treatment could activate tyrosine phosphorylation of cellular proteins and MAP kinase in a time and dose dependent manner. Using the dominant inhibitory S17N ras mutant, we show clear evidence that Ras is essential for HA-dependent activation of MAPK. Using an antisense oligonucleotide to FAK, we demonstrate that FAK is also required for MAP kinase activation. Treatment of HA with hyaluronidase abolished the activation of MAPkinase, confirming that MAPkinase activation was indeed triggered by HA. These findings are for the first time revealing critical steps in the HA signalling pathway .
