Several estrogen receptor (ER) variant mRNAs are present in various cell lines of human breast and endometrial cancers and their existence is considered to be one of the reasons for the loss of hormone dependence. The purpose of this study is to assess the ER variants in human various breast and endometrial tissues. The frequencies of three ER splice variants (del.2/3,del.5,and del.7) were assessed in 177 cases of breast and endometrial diseases by the reverse transcription-polymerase chain reaction (RT-PCR) method compared with the histopathological features. The expression of ER splice variants were not uncommon and almost of all cases showed one or more aberrant variants. Del.5 and 7 were more frequent than del.2/3 in both benign and malignant lesions. There was no significant difference between histo- pathological features, ER protein status, stage grouping and the frequency of each splice variant in cancer cases. We conclude that the ER splicing is a physiological phenomenon rather than an abnormal alteration in human breast and endometrial diseases.
